Sildenafilum
Active ingredient: Sildenafil (as sildenafil citrate) – 25 mg, 50 mg, or 100 mg.
Excipients: Microcrystalline cellulose, anhydrous dibasic calcium phosphate, croscarmellose sodium, magnesium stearate, Opadry® blue and Opadry® clear film coating.
Blue, film-coated, diamond-shaped tablets, slightly biconvex with beveled and rounded edges. One side is engraved with “Pfizer” and the other with “VGR 25”, “VGR 50”, or “VGR 100”, respectively.
Used for the treatment of erectile dysfunction, when the ability to achieve or maintain an erection sufficient for satisfactory sexual activity is impaired.
Sildenafil is effective only in the presence of sexual stimulation.
To avoid complications, sildenafil must be taken strictly as prescribed by a physician.
The recommended dose for most patients is 50 mg, taken approximately 1 hour before sexual activity.
Depending on efficacy and tolerance, the dose may be increased to 100 mg or reduced to 25 mg.
The maximum recommended frequency is once per day.
Mild to moderate (creatinine clearance 30–80 mL/min): No dose adjustment required.
Severe (creatinine clearance ≤30 mL/min): Dose should be reduced to 25 mg.
In patients with liver dysfunction, the dose should be reduced to 25 mg.
With ritonavir: The maximum dose is 25 mg, not more than once every 48 hours.
With CYP3A4 inhibitors (such as erythromycin, saquinavir, ketoconazole, itraconazole): The initial dose should be 25 mg.
With alpha-blockers: Sildenafil should only be used once hemodynamic stability is achieved; dose reduction may also be necessary.
No dose adjustment is required.
Hypersensitivity to sildenafil or any component of the formulation.
Concomitant use of nitric oxide donors, organic nitrates, or any form of nitrates is contraindicated due to the risk of severe hypotension.
Use with caution in patients with:
Anatomical deformities of the penis (including angulation, cavernosal fibrosis, Peyronie’s disease).
Conditions predisposing to priapism (such as sickle cell anemia, multiple myeloma, leukemia, thrombocythemia).
Active bleeding disorders or peptic ulcer exacerbation.
Hereditary pigmentary retinitis.
Heart failure, unstable angina, myocardial infarction, stroke, or life-threatening arrhythmias within the last 6 months.
Severe hypertension (BP >170/100 mmHg) or hypotension (BP <90/50 mmHg).
The product is not intended for use in children under 18 years of age.
